



{"id":25,"date":"2019-10-09T15:31:53","date_gmt":"2019-10-09T15:31:53","guid":{"rendered":"https:\/\/research.unl.edu\/annualreport\/2019\/?p=25"},"modified":"2019-10-20T14:32:52","modified_gmt":"2019-10-20T14:32:52","slug":"viral-discovery-could-aid-vaccines-treatments","status":"publish","type":"post","link":"https:\/\/research.unl.edu\/annualreport\/2019\/viral-discovery-could-aid-vaccines-treatments\/","title":{"rendered":"Viral Discovery Could Aid  Vaccines, Treatments"},"content":{"rendered":"\n<p class=\"wp-block-paragraph\">A virus\u2019s genetic loss can become its evolutionary gain.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">This surprising discovery by Nebraska virologists provides some of the clearest experimental evidence that losing a gene can improve evolutionary fitness \u2013 in this case, by helping a virus regain the ability to replicate.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">The team made the discovery after deleting the so-called B1 gene from the vaccinia virus. Because B1 helps silence a host\u2019s antiviral alarms and allows vaccinia to copy itself, deleting the gene threatens vaccinia\u2019s survival, said team leader Matt Wiebe, associate professor of veterinary medicine and biomedical sciences.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">But the researchers continued culturing the B1-free strain before sequencing its genetic code to gauge how it evolved. The strain responded by deleting a single base pair \u2013 a fundamental component of DNA \u2013 while leaving nearly 200,000 others untouched. Photo by Craig Chandler \/ University Communications<\/figcaption><\/figure>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cWe were expecting that the virus may adapt another gene to compensate,\u201d said Wiebe, a member of the Nebraska Center for Virology. \u201cWhat we found instead is that the virus adapted by inactivating another gene. It was as if, upon cutting one wire, the best way to fix the problem was to cut another wire.\u201d<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">That inactivation occurred in a gene, B12, whose purpose is largely unknown. Previously, when researchers elsewhere deactivated just the B12 gene, leaving all others intact, they found no effect on the vaccinia virus. Wiebe said the findings indicate that when B1 \nis absent \u2013 but only then \u2013 B12 actually inhibits viral replication by alerting the host\u2019s immune system.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Leveraging that knowledge to hijack viral machinery could further the development of vaccines and treatments.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cIt\u2019s very interesting to understand what viruses might teach us,\u201d said Wiebe, whose research was funded by the National Institutes of Health and the Fred &amp; Pamela Buffett Cancer Center. \u201cIf this virus has a mechanism of activating our immune system, can we eventually use that to inhibit the growth of other viruses?
